Investigation of Xylitol Effect on Sugar Metabolism by Chromatographic Analysis of Intermediates in Human Saliva

Abstract
Objective: The purposes of this study were to evaluate the mechanism of sugar metabolites by analysis of metabolic intermediates in sugar metabolic pathways.

Methods: The sugar intermediates (glucose-6-phosphate, fructose-1-phosphate, fructose-6-phosphate, and 6-p-gluconate) was analyzed by high-performance anion exchange chromatography with pulsed amperometric detection (PAD) with gradient elution of sodium hydroxide. The saliva was obtained from five volunteers. The intermediates are mixed in saliva (control) and xylitol added saliva experimental). The sugar phosphates and 6-p-gluconate in control and xylitol added saliva were analyzed to evaluate the increase or decrease as time elapsed.

Results: Sugar phosphate and 6-p-gluconate, the intermediates of sugar metabolism, are successfully analyzed with good linearity. The concentration of glucose-6-phosphate and fructose-1-phosphate was gradually decreased with time elapsed, and the concentration of fructose-6-phosphate and 6-p-gluconate showed a tendency to increase for a certain period of time. The xylitol added saliva could mitigate the increase of fructose-6-phosphate and 6-p-gluconate, while the concentration of glucose-6-phosphate and fructose-1-phosphate wasn¡¯t influenced by xylitol presence.

Conclusion: Xylitol can be effected the sugar metabolite, and alleviate the organic acid formation in saliva.
